General Presentation

The IBBMC is housing the Yeast Structural Genomics. Our initial pilot project concerned the structure determination of yeast proteins of unpredicted fold. Within this program we have set up a technological platform for the high throughput determination of 3 dimensional structures protein. This setup includes:

-  cloning and expression of proteins
-  purification and characterization of proteins
-  protein crystallization
-  3D structure determination by X-ray diffraction analysis
-  bioinformatics analysis and data management

In order to accelerate the pace, we have introduced an automatic approach for many of the steps of the complex road leading from gene to 3D structure. The ensemble of technologies is organized now in a platform that is open to the scientific community and that has acquired the label RIO. The following services are offered:

-  semiautomatic cloning and expression testing in bacterial hosts
-  purification and, if necessary refolding, of overexpressed proteins
-  crystallization screening of purified proteins or protein complexes
-  3D structure determination and structural analysis
-  bioinformatic analysis of the protein sequences
-  software for experimental data storage and browsing
